New Vegas with mods for maximum desert

Fallout is the obvious choice. Though New Vegas has the strongest "isolation" feel to it.
Personally find this quite a bit in S.T.A.L.K.E.R. Shadow of Chernobyl and especially Call of Pripyat.
Yume Nikki and its ilk (.flow, LCD Dem and many others) also invoke a feeling of isolation.

Uru.

There are several desert worlds. Even the ones that aren't desert have a sense of isolation. The setup is:

There's an ancient civilization called the D'ni. The civilization collapsed. Some modern people found the ruins and started a volunteer effort to explore, research, and maybe restore some of it. But the that effort ended when one of the people died in an accident.

So you, the player, wander around the ruins of a twice abandoned civilization.

Just thought I'd jump in to mention that this is part of the Myst series/universe and is fantastic. Also he's right about the general sense of isolation, though I don't think it was entirely intended as it was begun as a sort of mmo and some of the puzzles, maybe only two, require other people to complete but they're not necessary and only add cosmetic upgrades to your personal hub world.

I think you can also play most of the game as an out-of-th-box single player experience with Uru: Ages beyond Myst and the Path of the Shell expansion.
