You can probably read about the general plot line on the QB wiki, but no unless you're a professional weeb who has mastered moon runes, you'll only have a faint idea as to why all these big-breasted women are fighting. However, it definitely does help to have watched the series, if you know the characters, then you're not completely lost as to why some of them are friendly and others hostile.

If by standard fighting you mean like Tekken where you press buttons to make moves and stuff in real time, no. It's turn based. The OP video cuts out a lot of the gameplay and game mechanics. If you want to see how it works, I reccomend you youtube Queen's blade/gate spiral chaos
