Edna's VA dying didn't stop the show and they didn't have the brains to replace one of their major recurring characters. So most likely the death or quitting of one of the other main VA's ought to do it. Like someone who voices multiple characters aka Flanders' VA or one of the main family's VA. That ought to finally hit the nail on the coffin. For example. If one of the three main kids were to die they'd likely replace them with a Cousin Oliver type. Yes they would be that stupid since Simpsons is basically a franchise and they'd need to do whatever it takes to keep that dead horse fresh.

The Simpsons just has to stop reusing characters.
It's biggest weakness is they stuff each episode full of side characters that just go through their catch phrases for quick gags.
They need to get back to doing stories that develop over the entire course of the episode and have new characters that do new things. I imagine they don't do this because they want to keep using what voice actors they have, and reserve new characters for guest stars that can't really do decent voice acting.

Make the simpsons a sci-fi show about the USA entering total war with russia and china, and they conscript every able bodied person. In time, the whole simpson family is drawn into the war, and then they start killing russians. It should go on for like 3 seasons, until every simpson and every springfield citizen is killed off.
Lots of things could be done: Bart goes militia, the family is separated with Marge and Lisa going into supply, maggie getting put in daycare and the boys getting soldier duty, Homer gets fit… lots of storylines could be done.
The first few seasons should be the whole preparation and escalation, and mid-season 2 the nuclear hell gets unleashed. By the last part of the last season, they all get killed.
